Biochemical characterization of a novel L-Asparaginase with low glutaminase activity from Rhizomucor miehei and its application in food safety and leukemia treatment.
A novel fungal gene encoding the Rhizomucor miehei l-asparaginase (RmAsnase) was cloned and expressed in Escherichia coli. Its deduced amino acid sequence shared only 57% identity with the amino acid sequences of other reported l-asparaginases. The purified l-asparaginase homodimer had a molecular mass of 133.7 kDa, a high specific activity of 1,985 U/mg, and very low glutaminase activity. RmAs...

Introduction: L-asparaginase is an anti-neoblastic agent used in the chemotherapy of lymphoblastic leukemia. This enzyme is widely distributed among microorganisms, plants and animals, but microorganisms have proved to be a better alternative for L-asparaginase, thus facilitating its large-scale production. Actinomycetes are filamentous, Gram-positive bacteria widely distributed in the marine e...

Milk clotting enzyme (M CE) produced by: Rhizomucor miehei was purified and characterized. The enzyme was purified 220.29-fold with specific activity about 14444.2 U/mg protein byultrafiltration, ammonium sulfate fractionation, Sephacryl S-300 chromatography. The maximumenzyme activity was at 65°C.The milk clotting activity was decreased steadily as pH is increased and indicated maximumactivity ...
متن کاملProduction and characterization of biochemical properties of L-Asparaginase by indigenous yeast isolated from soil of Iran
Introduction: The use of L-asparaginase as a chemotherapeutic agent is an effective strategy for the treatment of leukemic lymphoblasts. Since the use of bacterial L-asparaginase causes side effects due to hypersensitivity reactions, search for new enzyme sources is one of the active research areas. In this study, the characterization of produced L-asparaginase by indigenous yeast strains was i...
متن کاملPurification and Characterization of Asparaginase from Phaseolus vulgaris Seeds
L-asparaginase from bacteria has been used in treatment of acute lymphoblastic leukemia. The aim of this study was to purify and characterize L-asparaginase from Phaseolus vulgaris seeds instead of microbial sources. L-asparaginase was purified to apparent homogeneity. The enzyme has molecular mass of 79 kDa. The purified asparaginase had very low activity toward a number of asparagine and glut...
